FRET with Spectral Imaging and Linear Unmixing - In FRET applications, spectral imaging can be considered a variation of the sensitized emission technique that relies on excitation of the donor alone, followed by acquisition of the entire emission spectrum of both the donor and acceptor fluorescence instead of capturing data in two independent channels. This technique is used for non-destructive inspection of devices with very small features such as those found in modern semiconductors. Also covered are various imaging modes, such as brightfield, oblique illumination, darkfield, polarized light, and DIC microscopy. Basic Microscopy Contrast Modes in Reflected Light Microscopy In its standard configuration, a typical reflected light microscope is readily equipped to examine amplitude absorption specimens using brightfield incident light. The matrix can be used as a scoring rubric to determine the degree to which students have correctly identified minerals in thin section and described and interpreted rock textures.

This is in contrast to bright or dark field microscopy, where the image contrast is due to transmittance or scattering. The primary challenge for light microscopists since van Leeuwenhoek's time has been to enhance the contrast between pale cells and their paler surroundings so that cell structures and movement can be seen more easily. Commonly, piezoelectric ultrasound transducers are employed. Upon leaving the specimen, the electron beam contains information about the structure of the specimen that is visually enhanced by the microscope. Live-cell imaging techniques now involved a wide spectrum of imaging modalities, including widefield fluorescence, confocal, multiphoton, total internal reflection, FRET, lifetime imaging, superresolution, and transmitted light microscopy. The resolution of X-ray microscopy lies between that of light microscopy and electron microscopy.
